Background

The Gene Editing and Mouse Model Core Facility is primarily tasked with producing genetically modified animals (GMA) for intramural researchers. In addition to GMA, the core collaborates with intramural researchers to develop genetically modified cell lines. In addition to genome editing projects, the core also offers a variety of services associated with research animals such as mouse colony importation and cryo-preservation/recovery of mouse lines. The GEMM Core staff also provides their expertise in troubleshooting issues in either previously made mouse/cell lines or on-going genome editing projects designed by intramural investigators and/or their extramural collaborators.

Services We Provide

  • Develop genome editing strategy and generate locus specific reagents for genome editing projects
  • Transfection of mouse embryonic stem cells with targeting vector and subsequent culturing, clonal expansion, and clonal screening
  • Microinjection of genetically modified ES cells into mouse blastocysts to generate chimeras and their subsequent breeding to identify germline transmission or chimeric analysis
  • Culturing of user-provided genetically modified ES cells for QAL/QC, and subsequent blastocyst micro-injection
  • Single-cell embryo micro-injection of CRISPR/Cas9 system to generate targeted genomic mutations
    • Note: Traditional random integration transgene projects are outsourced
  • CRISPR/Cas9-mediated genome editing in mouse ES cells and immortalized cell lines
  • Derivation of mouse ES cell lines from existing genetically modified mouse lines
  • Mouse embryo cryo-recovery and cryo-preservation
  • Assist in troubleshooting problems with previously generated genetically modified mouse lines
  • Long-term storage of frozen mouse embryos
  • In collaboration with CMB/ACUC, assist intramural investigators in developing efficient breeding strategy to optimize animal use and minimizing the animal cost for the institute
