Characterization of Protein Posttranslational Modifications

Mass Spectrometry Group

Posttranslational modification of proteins plays an important role in the regulation and activity of proteins. Thus, their determination is critical to understanding their role in disease mechanisms. The group is actively involved in developing and applying MS techniques to the characterization of protein phosphorylation states, identification of sites of phosphorylation, and characterizing the types of glycans and their heterogeneity on glycosylated proteins. Specific studies include:

  • Development of specific methodologies for enrichment and characterization of phosphorylated peptides using titanium dioxide
  • Application of top-down and middle-down approaches to characterization of posttranslationally or chemically modified proteins
  • Characterization of sites of glycosylation on proteins, e.g., HIV gp120 and on monoclonal antibodies (including a collaboration with Prof. M. Przybylski, Konstanz University, Germany)
