Box 12233 Mail Drop F0-04 Research Triangle Park, North Carolina 27709 Delivery Instructions My research is focused on the use of hydrogen/deuterium (H/D) exchange and mass spectrometry to identify protein:protein interaction surfaces. This strategy involves exchanging the protein’s backbone amide protons with deuterium and measuring the extent of deuterium uptake using mass spectrometry. Since protein:protein interactions generally involve the exclusion of solvent along the interaction surface, the rate of H/D exchange is slower along the surface area of the interaction, resulting in a decrease in deuterium uptake in the interaction region. The H/D exchange reaction is performed in the presence and absence of the protein’s interaction partner, and the extent of deuteration is determined by digesting the protein and analyzing it using mass spectrometry. Peptides that exhibit a reduction in deuterium uptake in the presence of the interaction partner are identified using tandem mass spectrometry and evaluated for their potential involvement in the interaction surface. .
